Problem -
After opening an AVS, if I tried to open another AVS (a new file), VDUB would crash. It wouldn't matter what filters or what not was in the second or first AVS. It didn't matter which version of VDUB I had (I tried 1.50+). It didn't matter what version Avisynth I had (I tried 2.5+).
Solution -
I simply deleted the registry information using REGEDIT out of my system for Avisynth. Essentially deleting the path to the plugin dir. So now I have to manually have an LoadPlugin line in my scripts, but VDUB no longer crashes :D
Hope this helps some folks. Oh, and I also re-installed XP and on a fresh install I was having this issue too, and the same solution worked. So... it might be a bug.
