1. Do matroska chapters need to be on a keyframe? If they aren't, would it seek to the nearest keyframe, or would it start playback from the actual time that was specified?
2. How do you actually use the chapters on playback? I tried playing in media player classic and the core media player, and in neither one was I able to figure out how to access the chapters

It does not need keyframes. But it does not hurt. Keyframes are the most efficient and easy to use.

In MPC page down goes to the next chapter and page up goes to the previous chapter.

Apparently something has gone wrong then, because this is not working. If I open up the file in vdubmod, it lists the chapters, however not in mpc. Any ideas?

Yes but remember that only mkvtoolnix is able to insert chapters. With virtualdubmod i have problems (they don't appear).

VirtualDubMod only mux chapters into OGM, not MKV at the moment. I will also love this feature and attachement into VDMOd, but let's see if in the next build my dreams come true... :p

VirtualDubMod only mux chapters into OGM, not MKV at the moment. I use VdubMod 1.5.4.1 (DVDtoOgm) for creating mkv and there are chapter in my files ;)

You should see the following thread:

It concerns MPC and all its wonders (and remaining problems for some).
