Can vspreview use other sources like DirectShowSource? If so, how?

Your questions (except those about placeholders) are not related to vspreview, those are vapoursynth questions, related to scripts. vspreview in your title questions is kind of screwing up searches and newbies as well.

Whatever works as in a vaporsynth script, vspreview would play it. If script does not output correct vs.VideoNode because there is an error in a script (like wrong source plugin, syntax error etc.), vspreview would not play it.
Not sure why DirectShowSource is ever needed nowadays. I'd use other sources. It might have problem with frame accuracy.
