I have here a folder that a contains a full Blu-Ray rip. There's a folder called BDMV and a second one called CERTIFICATE. How can I make an image (.iso or something) from this with a Mac? For Windows there's ImgBurn with it's fantastic 'Build'-Mode, is there something comparable on MacOSX or can it be done with built in DiskUtility?

You need a burning application that supports UDF 2.50. But I don't know if macosx has udf 2.50 support.

Maybe this helps:

go and grab toast titanium, there is a burning option für bdmv folders ;) works like a charm. Or you can create an image file too....but for what do you need iso?
