I've created a custom SubMenu via DVDLabPro.
4 options
On
Off
Main Menu
Play Movie
If I select On, then select play movie, I don't get subtitles that appear, unless I use my remote or in VLC player, right-clicking to turn them on.
Please someone help...Thanks.

Probably something in your title that turn them off. Trace the DVD through with PgcEdit to see what happens exactly.

Did you? If you did, you woud have seen the last command in title 1, which (re)sets the subpic to the value in gprm(12).
If, in your subpic menu, instead of setting sprm(2) to 64, you set gprm(12) to 64, you should be OK. Similarly, to turn it off in your subpic menu, set gprm(12) to 0.
The pre commands of the subpic menus (PGCs 14 and 15 - well, PGC 15 is irrelevant since is it unreferenced) are not well constructed - since they reset gprm(12). You need to test for whether the subpic is on or off by comparing the value in gprm(12) to 64 and then picking button 2 or 3.

Maybe the easiest would be to delete the last line in the title. See how you go with that.

@EuropeanMan
Hi!
What happens if you select "On" and then select "Main Menu" - and then select "Play Movie" from the Main Menu screen?
