The standards are different in the delivery modulation/coding scheme but both carry MPEG-2 transport. If you have only files, then you have only the MPEG-2 payload and so may not be able to distinguish them. If they use different program guide data schemes, or any other different metadata, you may be able to distinguish them that way.
Why do you need to distinguish them?

Thanks for the reply.
My query is:
If I have four DVB Transport streams with SI information and i need to know what kind of modulation is used with these files then where i need to check for that in which section i need to know, i agree this information is carried in NIT (used to specify physical network characteristics).

You can look in the NIT for the presence of these descriptors, which contain modulation info:
cable delivery system descriptor
satellite delivery system descriptor
terrestrial delivery system descriptor
The mere presence of the descriptors may tell you what you want without having to look at the modulation, i.e., if you have a cable descriptor then you have DVB-C etc.
