In general not for a 3.5", could work with a 2.5" YMMV

Even with a 2.5 inch hard disk, generally two USB ports are required. One for power/data, the other just for power. A non powered hub will NOT work (if it does I wouldn't rely on it).

USB isn't designed to provide power, but it can. There's heaps of gimmicky devices designed to run off USB power but most don't work well because of such little power.

you can pick up a powered USB hub for about 15-40$ and just plug it in behind the home theater system, for power and just plugging into the player with the data port

we used to run portable usb harddrives off of one usb port all the time in school, but the boards we were working with were entery level server Mobo, with he laptops we got reduced speed using only one port though

Just be careful with some of the powered USB hubs out there, many powered hubs can't supply what they claim they can.
