No, it doesn't. DivX 5 does not support multicore at all. Only VDubMod (which is used to encode) puts AviSynth into an extra thread that runs on the second core. You'll only notice a difference with very CPU intensive scripts though.

Bottom line is: To push a quad core CPU to its limits with DivX 5 you need to run 3-4 encodings simultaneously. Might get you into trouble with too little RAM though.
