A Clarification

I just read through the thread regarding the audio /video sync.

It may be stupid question, but I just wanted to ask the members, if the video in question must have an audio muxed in or the without the audio.

The thread in question refers to video with multiplexed audio - How else would you know that the audio and video are or aren't in synch?
