I have installed and uninstalled avisynth sucessfully the first time. However, I am unsucessful on my next try with the error message:
Could not copy avisynth.dll to system directory-Close down all applications that use Avisynth, and sure to have write permission to system directory, and try again.
I have admin rights and there were no other programs running.
I have also ensured that none of the registry keys below is there when I tried installing again but I still get the same error message.

I don't have any idea why, but this happened to me a while back and I couldn't get it unstuck for any reason. So I moved avisynth.dll out (not sure if you can in explorer, I used cmd) to c:\ and it was suddenly deleteable. Played with permissions, made sure nothing had a handle to it, rebooted, all kinds of things, only moving it worked.

But I don't have avisynth.dll anywhere on my harddisk...so I can't move it around or delete it.

Exactly the same thing happened to me. When I tried to reinstall avisynth, after uninstalling it, I got the same message as you.
Have you recently installed Microsoft.NET Framework 2.0? This particular program corrupted AVS Edit 1.1.1.1, although at first I thought that the problem lay with avisynth. Hence my reason for uninstalling it.
I have two machines, and fortunately NET Framework 2 was only installed on one of them. I managed to "get" avisynth on the problem machine by copying the avisynth dll from the clean machine to the problematic one, and similarly I copied the avisynth folder, the one that exists in the Program Files, to the corresponding Program Files of the problem machine. Then I ran the avisynth executable and everything now is hunky dory.
Of course if you don't have a second machine, or it's not appropriately set up, this is not a way out for you.

I had the same problem, i installed avisynth on another computer, copied the dll from the other computer system32 to my computer system32 folder and then i was able to install it normally.

after installing avisynth so, I need to restore the modified dll for the usage of .netframework2?

I had the problem but I kept reinstalling it, and it worked one time. That was after I deleted the Program Files folder for avisynth.

Please install AviSynth 2.5.7 Alpha2 [April 2nd]- DIAG Release! (http://forum.doom9.org/showthread.php?t=109477). The installer in this release has been doctored to give much more diagnostic information on this type of failure.
If you do get a failure please take a screen grab of the message. Also please arrange the 2 windows so they are not overlapping before you snap the screen, all the information in both windows could be relevant.
Thanks in anticipation for your help in resolving this problem.

Avisynth 2.57 is definitely compatible with megui. I only know of a few plugins that need SetLegacyPlanarAlignment, and one that simply crashes it (mpeg2dec3), which works with 2.56. All of the ones megui uses are compatible with 2.57.

If you want the installer problem resolved, then please run the special 2.57 Alpha 2 installer and report exactly what happens. I cannot fix what I cannot see! Since I put the special diagnostic version up I have had only 1 active response, I am a little disappointed.
You can always re-copy neutrogenik's posted avisynth.dll if you have problems with the 2.57 version, but I would not expect you to have any conflict problems with megui.

I saw this problem on another forum too. I think (as was the case there and sometimes here) the only problem is that avisynth.dll is sometimes set on read only, and thus not overwriteable.
