If you encode with CCE then you should leave convert to YUY2() on as CCE only takes input of YUY2 color space.
When HC or Quenc then you should leave it off because they take input of YV12. But don't worry jdobbs has made DVD-RB inteligent and it will automatically choose the best option for you even if the source is interlaced.
The above info. I found by 'onesoul'.
However, I didn't really find any info. to indicate that I should untick the 'convert to YUY2() off when using the HC encoder in the help section of DVD-RB Pro.
I have been leaving Convert to YUY2() engaged even while using DVD-RB Pro. in HC mode, Should I? Does it matter? Will DVD-RB automatically choose for me?
