The .sup format contains the subtitles as bitmap images (just like the original subtitles on the DVD).
The .srt format contains the subtitles as text.
So you need to run the .sup file through an OCR program. DVDSubEdit can do it for you. It's definitely worth a try, but the result depends very much on how your bitmap subtitles look (font style? Are there any italic subtitles mixed with subtitles in normal formatting? Etc.)
If you get too many unrecognized characters, you should try SubRip instead. SubRip lets you "teach" the OCR engine to recognize the characters in your subtitles, as you can add information to the program's "character matrix". It takes longer time but you will be able to get an accurate output.
However, IIRC, SubRip can't read .sup files, so you'd need the original DVD (where you got the .sup file from) - or create/mux a temp. DVD with your .sup file to be loaded in SubRip.
